What would happen if you use Spidermans Special Attack against a Venoc Viper when you mix them? I don't see what could happen with -1 def.

Nothing, nothing would happen.

You roll 1 less Defense Die. One less than nothing is still nothing not -1. You don't unroll a defense die or anything like that.

If they got one defense die for elevation, Thorgrim, Gerda, etc., they would lose it. Jalgaurd glyph and Raelin would be half as effective for them.

Yep, nothing will happen. But Id like to think of it as Amplify Damage (one of Slitherin Guard's ability in DOTA). For every negative defense die, Spidey adds 1 attack die. I said "Id like to think of it as" but wont really do it in a game.
